Angiotensin-converting enzyme (ACE) is a zinc-containing dipeptidyl carboxypeptidase widely distributed in mammalian tissues and plays a critical role in blood pressure regulation. The ACE precursor consists of 1306-amono acid residues including a 29 amino acid residue-signal peptide. The mature protein has a molecular weight of 180kDa. There are two isoforms of ACE, a smaller/ single catalytic site enzyme found in the testes and a more widely expressed ~180kDa dual active site isoform. ACE is often membrane bound.² ACE inhibitors are widely used to treat hypertension and cardiovascular diseases. ACE inhibitors have also been used to slow nephropathy, particularly when it is associated with diabetes.³ The standard product used in this kit is recombinant human ACE, consisting of 30-1261 amino acids with the molecular mass of 120 kDa.
