The Cytofect™ Preadipocyte Transfection Kit is a plasmid DNA delivery system optimized specifically for primary preadipocytes. Primary preadipocytes can be notoriously difficult to transfect without inducing cytotoxic stress or disrupting their adipogenic differentiation potential. The Cytofect™ system is engineered to support efficient nucleic acid delivery and high cell viability in primary precursor models before, during, or after differentiation.

Preadipocytes are fibroblastic, mesenchymal precursor cells residing within the stromal vascular fraction (SVF) of adipose tissue. The kit is compatible across primary preadipocyte models from diverse anatomical depots:

  • Subcutaneous & Visceral Depots: White adipose tissue (WAT) precursors involved in systemic lipid storage, energy homeostasis, and metabolic regulation.
  • Thermogenic Depots: Brown adipose tissue (BAT) and beige precursor populations capable of acquiring thermogenic capacity under specific biochemical cues.
Through adipogenesis, preadipocytes undergo a regulated transcriptional cascade to differentiate into mature adipocytes. As they mature, cells transition from a fibroblastic morphology to lipid droplet-accumulating cells with specialized metabolic and endocrine roles:

  • Energy Storage & Mobilization: Regulating triglyceride synthesis, lipolysis, and insulin-stimulated glucose uptake.
  • Endocrine Signaling: Secreting key adipokines, cytokines, and growth factors that regulate systemic insulin sensitivity, appetite, and inflammatory pathways.
  • Thermogenesis: Expressing uncoupling protein-1 (UCP1) in brown and beige adipocytes to drive adaptive thermogenesis and energy expenditure.
The kit provides a complete reagent system containing Cytofect-2, an endosomolytic Enhancer, transfection medium, and antibiotic-free growth medium. The Enhancer complexes with plasmid DNA and Cytofect-2 to promote cellular uptake and endosomal escape, facilitating target plasmid transport toward the nucleus.

Plasmid DNA delivery into primary preadipocytes enables transient overexpression, reporter construct assays, and targeted gene functional studies. Key research applications include:

  • Transcriptional Regulation of Adipogenesis: Investigating master regulators (e.g., PPARγ, C/EBP family) during early and late lineage commitment.
  • Metabolic Disease Modeling: Examining insulin resistance, lipid droplet dynamics, and altered adipokine secretion in models of obesity, Type 2 diabetes, and metabolic syndrome.
  • Inflammatory Signaling: Analyzing crosstalk between preadipocytes, hypertrophic adipocytes, and infiltrating immune cells during tissue remodeling.

This kit contains Cytofect-2, Viral Enhancer* and Media for transfection and culturing cells. Viral Enhancer complexes with DNA and Cytofect-2 and escorts the transfection complex to the nucleus. This results in high transfection efficiency and viability for these traditionally hard-to-transfect primary cells.

The Kit contains all necessary reagents, media and optimized protocols to carry out successful transfection of Preadipocytes with good efficiency (25%) and viability (80%-95%).
