The Cytofect™ Mesenchymal Stem Cell Transfection Kit is a plasmid DNA delivery system optimized specifically for primary mesenchymal stromal/stem cells (MSC). Primary MSC can be notoriously difficult to transfect without inducing cytotoxicity or triggering premature differentiation. The Cytofect™ system is formulated to achieve efficient plasmid delivery and high cell viability while maintaining the multipotent phenotype of cultured primary stem cells.

Mesenchymal stromal cells — frequently referred to as marrow stromal cells when isolated from bone marrow — are multipotent, non-hematopoietic progenitor cells residing within connective tissue and perivascular niches. Primary MSC are routinely isolated from diverse tissue sources, including:

  • Somatic Niches: Bone marrow aspirates, adipose tissue, dental pulp, and synovial membranes.
  • Perinatal Niches: Umbilical cord matrix (Wharton’s jelly) and placental tissues.
Primary MSC populations exhibit natural heterogeneity influenced by tissue origin, donor demographics, isolation protocols, and culture passage numbers. In native tissues, MSC and perivascular stromal cells establish microenvironmental niches that regulate tissue homeostasis, vascular architecture, and crosstalk with immune and hematopoietic cells.

In culture, primary MSC maintain the capacity to proliferate and differentiate along three principal mesenchymal lineages:

  • Osteogenic Lineage: Generates functional osteoblasts to model bone formation, matrix mineralization, and metabolic bone remodeling.
  • Chondrogenic Lineage: Differentiates into chondrocyte-like matrix-producing cells for cartilage repair and joint biology studies.
  • Adipogenic Lineage: Forms mature adipocytes to investigate lipid metabolism, adipokine secretion, and mechanisms of insulin resistance.
Beyond structural differentiation, MSC perform vital paracrine functions by secreting bioactive factors, extracellular vesicles, and immunomodulatory cytokines that influence cell survival, angiogenesis, and inflammatory responses.

The kit provides a complete system containing Cytofect-2, an endosomolytic Peptide Enhancer, transfection medium, and antibiotic-free growth medium. The peptide Enhancer complexes with plasmid DNA and Cytofect-2 to promote endosomal escape and target the transfection complex toward the nucleus.

Plasmid DNA delivery into primary MSC enables transient overexpression, gene knock-in, reporter assays, and pathway modulation. Key downstream applications include:

  • Stem Cell Biology & Lineage Commitment: Investigating transcription factors regulating osteogenesis, chondrogenesis, and adipogenesis.
  • Immunomodulation & Niche Dynamics: Modeling MSC crosstalk with hematopoietic cells, immune populations, and leukemic microenvironments.
