The Cytofect™ Chondrocyte Transfection Kit is a plasmid DNA delivery system optimized for transfection of primary human chondrocytes, including articular chondrocytes isolated from diarthrodial joints. Primary chondrocytes can be difficult to transfect while maintaining viability and characteristics associated with their native phenotype. The kit contains Cytofect-2, Viral Enhancer, transfection medium, and antibiotic-free growth medium. The Viral Enhancer is an endosomolytic peptide formulation that complexes with DNA and Cytofect-2 and promotes delivery of the transfection complex toward the nucleus. This formulation is designed to support plasmid delivery, cell viability, and transgene expression in primary chondrocytes, with approximately 35% transfection efficiency and 80–95% viability for human chondrocytes under specified conditions; actual results may vary with cell source, passage, plasmid, and experimental conditions.

Chondrocytes are the principal resident cells of cartilage. They are found in specialized tissues including articular cartilage covering the surfaces of synovial joints, the intervertebral discs, the nasal septum, and the tracheal rings. Articular cartilage is an avascular, generally aneural tissue in which chondrocytes are embedded within a dense extracellular matrix. Although the articular surface contacts synovial fluid, chondrocytes are surrounded by cartilage matrix and receive nutrients primarily through diffusion. During development, chondrocytes arise through the chondrogenic differentiation of mesenchymal progenitor cells. In culture, primary chondrocytes may proliferate and can undergo phenotypic changes, including loss of their rounded morphology and reduced expression of cartilage-associated genes with extended expansion.

The primary function of chondrocytes is to synthesize, organize, and maintain the cartilage extracellular matrix. In articular cartilage, this matrix contains abundant type II collagen and aggrecan. The collagen network provides tensile reinforcement and structural organization, while aggrecan-associated proteoglycans help retain water and contribute to resistance against compressive loading. Chondrocytes also regulate matrix turnover by producing matrix components and remodeling enzymes and by responding to mechanical, inflammatory, and biochemical signals.

By enabling plasmid DNA delivery into cultured primary chondrocytes, the Cytofect™ Chondrocyte Transfection Kit supports research into cartilage development, extracellular matrix synthesis, inflammatory signaling, mechanotransduction, and chondrocyte phenotype. Transfected chondrocyte models can be used to investigate gene function and gene regulation, examine pathways involved in cartilage degradation and repair, and evaluate candidate therapeutics for osteoarthritis and other joint diseases. These models can also support regenerative medicine research and the development of experimental approaches that are subsequently evaluated in three-dimensional cultures, explants, animal models, or other translational systems. The kit is intended for research use only and is not intended for human, animal, or diagnostic applications.
