Cytofect™ Endothelial Cell Transfection Kit is a specialized transfection reagent system designed to support the delivery of nucleic acids into cultured endothelial cells. The kit can be used with several endothelial cell type models, including primary human aortic, pulmonary artery, umbilical vein, coronary, and microvascular endothelial cells, as well as appropriately validated endothelial cell line models.

Cytofect™ Transfection Kits are specifically optimized for transfecting primary cells, all in one complete, affordable system. Other commercially available transfection kits are optimized on immortalized cells.

Endothelial cells form the inner lining of blood vessels and are found throughout the body within the vascular network of tissues and organs, including arteries, veins, capillaries, and specialized microvascular beds. Aortic and coronary endothelial cells contribute to the regulation of large-vessel and cardiac blood flow, pulmonary endothelial cells regulate gas exchange and barrier function, umbilical endothelial cells provide a model of fetal and placental vascular biology, and microvascular endothelial cells participate in tissue-specific exchange and vascular remodeling.

In vivo, endothelial cells regulate vascular permeability, platelet interactions, leukocyte trafficking, coagulation, angiogenesis, and the delivery of oxygen and nutrients to tissues. They also respond to inflammatory mediators, shear stress, hormones, and hypoxic signals. Because endothelial dysfunction contributes to cardiovascular disease, inflammation, cancer, diabetes, and impaired wound healing, cultured endothelial cells are widely used in basic, translational, and clinical research.

Cytofect™ is formulated to support efficient delivery of plasmid DNA and other compatible nucleic-acid cargoes into endothelial cells while helping preserve cell viability. The system may be used for transient gene expression, reporter assays, promoter studies, protein production, pathway analysis, and functional studies of angiogenesis, inflammation, barrier integrity, and vascular signaling. The kit may also support experiments involving gene knockdown or, when paired with an appropriate editing system, gene editing.

The kit’s components are intended to simplify optimization of an endothelial transfection protocol. The kit components include the transfection reagent and a peptide enhancer or endosomolytic peptide designed to assist intracellular delivery. Researchers should consult the current kit components list and product documentation for the exact kit components, recommended reagent-to-nucleic-acid ratios, incubation times, and compatible culture formats. Because individual applications vary due to differences in tissue site, donor, history, nucleic acid type, and purity, optimizing conditions for each experiment helps ensure the most reliable results.

Endothelial transfection models can be used to investigate tumor–vascular interactions, including studies involving tumor cells, angiogenic signaling, vascular permeability, and the endothelial response to cancer-associated factors. They may also contribute to research relevant to cardiovascular disease, pulmonary vascular biology, reproductive health, and vascular complications associated with disease. Applications related to cancer screening and diagnostic procedures generally require additional disease-specific samples, validated assays, and clinical workflows;

Cytofect™ Endothelial Cell Transfection Kit is intended for research use only. It is not intended for direct clinical administration, diagnostic procedures, cervical cancer screening, or therapeutic use in humans or animals. The product should be qualified for each endothelial cell source and application by measuring transfection efficiency, viability, morphology, and the intended molecular or functional endpoint.
