Preadipocytes—alternatively designated as human primary preadipocytes or adipose precursor cells—are committed, fibroblast-like progenitor cells residing within the stromal vascular fraction (SVF) of adipose tissue. While multipotent mesenchymal stem cells retain the capacity to differentiate into various skeletal lineages, preadipocytes have restricted their developmental potential exclusively to the adipogenic lineage (adipogenesis). In their undifferentiated state, these cells lack unilocular lipid droplets and active adipokine secretion networks, instead functioning as a highly proliferative reservoir that expands and differentiates in response to systemic metabolic and endocrine cues.

By utilizing human primary preadipocytes alongside human induced pluripotent stem cell (HiPSC)-derived lineages, investigators can model the dynamic mechanisms of adipose expansion. Preadipocyte models are essential tools for mapping the distinct molecular pathways governing white versus brown adipose differentiation, characterizing line-specific metabolic defects, exploring tissue-specific inflammation cascades, and executing high-throughput screening for anti-obesity and insulin-sensitizing drug candidates.
