Canine Aortic Smooth Muscle Cells (CnAOSMC) are primary smooth muscle cells derived from the tunica intima and tunica media of normal canine aorta, serving as a well-characterized experimental model in vascular biology and cardiovascular research.

In vivo, vascular smooth muscle cells populate the arterial wall, where they regulate vascular tone, structural remodeling, and hemodynamic properties. In experimental research, CnAOSMC — alongside related mammalian models such as human primary cells and rat primary cells — can be used to investigate mechanisms relevant to vascular disease, arterial disease, and vascular smooth muscle cell function. For instance, studies using CnAOSMC have demonstrated that platelet-derived growth factor (PDGF) acts as a potent mitogen and chemotactic agent involved in intimal hyperplasia and atherosclerosis. Research has revealed significant regional differences in PDGF production within the normal canine aorta, with smooth muscle cells contributing to this variation, whereas graft smooth muscle cells often exhibit altered functional properties, including increased PDGF production that may promote intimal hyperplasia. Furthermore, these cells have been utilized to evaluate how agents like nebivolol relax vascular smooth muscle through nitric oxide- and cyclic GMP-dependent mechanisms, and to identify cDNA for the canine isoform of nitric oxide synthase (iNOS) to study allograft rejection and vascular pathology.

Beyond smooth muscle physiology, canine primary cells and stable cells provide valuable platforms for broader biomedical investigations, encompassing cellular analysis, molecular weight characterization of signaling proteins, ligand-binding capacity assays, and studies involving mesenchymal stem cells, vascular SMC populations, and calcium channels. Maintaining these cultures requires careful adherence to standard sterile technique and consistent culture conditions utilizing appropriate growth medium and cell culture reagents to support rigorous experimental analysis.

Canine Aortic Smooth Muscle Cells (CnAOSMC) are derived from tunica intima and tunica media of normal canine aorta. They are cryopreserved at second passage and can be cultured and propagated at least 10 population doublings.

A sampling of studies using CnAOSMC concluded the following:

  • Platelet-derived growth factor (PDGF) is a potent mitogen and chemotactic agent which may be involved in intimal hyperplasia and atherosclerosis
  • Significant regional differences in PDGF production exists in normal canine aorta, and that SMC are a significant contributor to the regional variation in PDGF production
  • Nebivolol relaxes vascular smooth muscle by NO- and cyclic GMP-dependent mechanisms
  • Graft SMCs are functionally altered producing more platelet-derived growth factor (PDGF) than aortic SMCs. PDGF produced by graft SMCs may contribute to the development of intimal hyperplasia. Graft SMCs have decreased proliferation response, but have similar migratory response to PDGF compared with aortic SMCs
  • Intrinsic differences in endothelial cells from proximal aorta versus the distal aorta have different capacity to produce PDGF in response to stimulants whereas unstimulated SMCs do not exhibit regional variation in PDGF production and did not increase PDGF secretion after PMA or thrombin treatment
  • Identification of the cDNA for canine isoform of nitric oxide synthase (iNOS) can be used in the study of allograft rejection and cardiovascular disease
