Human Chondrocytes: Re-Differentiated (HC-RD) — also known as mature chondrocytes — are primary cells that have undergone a controlled, multi-step rescue process to restore their native phenotype. When articular chondrocyte populations are expanded in two-dimensional (2D) monolayer culture to generate sufficient biomass, they transition into dedifferentiated chondrocytes, losing their specialized function. To reverse this, investigators encapsulate passaged chondrocytes into a three-dimensional (3D) suspension environment, such as chondrogenic medium or specialized chondrocyte medium. This spatial restriction provides the cues necessary for the cells to regain their native spherical morphology, re-establish chondrogenic differentiation, and reactivate the synthesis of cartilage matrix components, specifically Type II collagen (or II collagen) and aggrecan.

The primary utility of HC-RD lies in their ability to regain the functional characteristics of native human articular cartilage.

  • Transcriptional Reprogramming: Upon 3D encapsulation, HC-RD upregulate SOX9, the master transcription factor driving the production of collagen type II. Researchers carefully monitor the differentiation index—the ratio of type II to type I collagen—to ensure the population has successfully suppressed markers of fibrocartilage or chondrocyte hypertrophy typical of hypertrophic chondrocytes.
  • Quality Control: Unlike mesenchymal stem cells (or a mesenchymal stem cell lineage) which require extensive directed induction, HC-RD represent a specialized lineage. While they effectively produce a cartilage matrix, researchers must balance factors such as chondrocyte proliferation and chondrocyte maturation to ensure the resulting tissue mimics the biomechanical properties of hyaline cartilage.
HC-RD serve as a premier platform for studying the molecular checks and balances that govern cartilage health. In biomedical engineering and research, they are frequently benchmarked against other models, such as mesenchymal stem cells, synovial fibroblasts, or immortalized chondrocyte cell lines (e.g., C28/I2).

  • Mapping Catabolic Cascades: In models of osteoarthritis, HC-RD are challenged with proinflammatory cytokines (e.g., IL-1β, TNF-α). Researchers measure the suppression of potent matrix-degrading enzymes—such as MMP-13 and ADAMTS-5, which are key drivers of structural joint erosion—and monitor for signs of chondrocyte apoptosis to validate potential chondroprotective therapies.
  • Regenerative Pipelines: Because human cartilage possesses restricted innate repair capacity, HC-RD are essential for autologous chondrocyte implantation and 3D bioprinting strategies. By tracking cellular integration into a cartilage lesion and observing the effects of specific growth factors, researchers aim to optimize protocols for cartilage regeneration and cartilage tissue engineering, providing a reliable foundation for advancing new osteoarthritis treatments.
Human Chondrocytes Re-Differentiated (HC-RD) have been encapsulated in an alginate bead suspension.  HC-RD exhibit spherical morphology and have regained their ability to produce the phenotypic markers Aggrecan and Type II Collagen.  RD-HC can be used in tissue engineering experiments aimed to repair cartilage defects and to study chondrocyte differentiation and the effects of Growth factors & Cytokines on the suppression and maintenance of differentiated cartilage phenotype.

Instructions to release HC-RD from Alginate Beads

  • Remove Chondrocyte Differentiation Medium
  • Wash the beads 2 times with 0.155 M NaCl
  • Add 3 volume of Depolymerization Solution per volume of packed alginate beads.
  • Mix at room temperature for 30 minutes with gentle agitation until the beads have completely dissolved
  • Pellet re-differentiated chondrocytes at 1000 rpm for 10 minutes
  • Wash the cells 2 times with 0.155 M NaCl
Media Salts & Chemicals used to Release HC-RD from alginate beads:

  • Calcium Chloride Solution: Formulated to induce polymerization of Sodium Alginate.
  • Sodium Alginate:  Encapsulates chondrocytes to maintain their phenotypic characterization after isolation or to induce cultured chondrocytes to re-differentiate to chondrocytes
  • Depolymerization Solution (DS): Consists of sodium citrate, and dissolves alginate beads to release re-differentiated chondrocytes.

Cells shipped 10 days after encapsulation.  Re-Differentiated Chondrocytes expected 3 weeks after encapsulation, retain phenotype at least 1 week after complete re-differentiation.

Chondrocytes cultured in monolayer quickly lose their phenotypic characteristics and no longer express Collagen type II and sulfated proteoglycan. Encapsulation of de-differentiated chondrocytes for 3 to 4 weeks in alginate beads will induce re-differentiation and ECM production in chondrocytes.
  • Calcium Chloride Solution is formulated to induce polymerization of Sodium Alginate.
  • Chondrogenesis Kit contains all media & solutions for the encapsulization of chondrocytes in alginate beads
  • Sodium Alginate encapsulates chondrocytes to maintain their phenotypic characterization after isolation or to induce cultured chondrocytes to re-differentiate to chondrocytes.
  • Depolymerization Solution (DS) consists of sodium citrate, and dissolves alginate beads to release re-differentiated chondrocytes.
Instructions for Releasing Re-differentiated Chondrocytes from Alginate Beads

  1. Remove Chondrocyte Differentiation Medium
  2. Wash the beads 2 times with 0.155 M NaCl
  3. Add 3 volume of Depolymerizatiojn Solution per volume of packed alginate beads
  4. Mix at room temperature for 30 minutes
  5. Apply gentle agitation until the beads have completely dissolved
  6. Pellet re-differentiated chondrocytes at 1000 rpm for 10 minutes
  7. Wash the cells 2 times with 0.155 M NaCl
