Description

BACKGROUND Chemokines (chemotactic cytokines) are small heparin-binding proteins, which constitute a large family of peptides (60-100 amino acids) structurally related to cytokines whose main function is to regulate cell trafficking. They are subdivided into four families based on the number and spacing of the conserved cysteine residues in the N-terminus of the protein: CXC, CC, CX3C, and C, in agreement with the systematic nomenclature. Chemokines play a major role in selectively recruiting monocytes, neutrophils and lymphocytes as well as inducing chemotaxis through the activation of G-protein-coupled receptors (GPCR).1

MIP-1α is a member of the C-C chemokine family. Similar to MCP-1, MIP-1α can mediate the recruitment of monocytes in several inflammatory diseases. MIP-1α also leads to a local accumulation of neutrophils when it is subcutaneously injected. MIP-1α has been shown to play a role in several inflammatory diseases. For example, bleomycin-induced pulmonary fibrosis is decreased when MIP-1α is inhibited, and influenza-induced pneumonitis is dependent on the presence of MIP-1alpha. These many studies support the notion that both MCP-1 and MIP-1α are key molecules in macrophage trafficking into inflammatory foci.2 In addition, it was shown that MIP-1alpha attract CD8+ T cells, B lymphocytes, and NK cells.3 It can stimulate basophil to release histamine and induce ICAM-1 expression, mast degranulation, and production of TNF-alpha, IL-1, and IL-6.2

MIP-1α binds to CCR1, CCR4 and CCR5 chemokine receptors. It is one of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1α induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV).4
